Synergy for Salmon: Study Spawns Insight into Pesticide Mixtures

چکیده
Organotins have been widely used as agricultural pesticides, anti fungal agents, polyvinyl chloride stabilizers, industrial catalysts, and antifouling additives in boat paints. These tinbased chemicals, which have been detected in various environmental media, are lipophilic and thus capable of becoming increasingly concentrated as they pass up the food chain. A new study suggests that the toxic effects of organotins on living cells are mediated in part by inhibiting the function of the proteasome, a molecular structure that degrades unneeded or damaged proteins [EHP 117:379–386; Shi et al.]. In eukaryotic organisms (animals, plants, fungi, algae, and plank ton), more than 80% of intracellular proteins are degraded through the proteasomemediated pathway. By interfering with proteasome function, organotins enable proteins to accumulate inappropriately. Because normal immune function and many cellular processes depend on the proteasome pathway, the organotin–proteasome interaction could help explain some of the adverse health effects of organotins— notably endocrine disruption, infertility, and immune dysfunction— that have been observed in wildlife and in animal studies. In addition, human exposure to organo tins has been proposed as a possible risk factor for cancer (by inhibiting the cytotoxic function of natural killer cells), neurotoxicity, obesity, allergies, asthma, and altered reproduc tive development. The researchers provide several lines of evidence suggesting that triphenyltin (TPT), a common organotin, binds to and blocks the activity of the proteasome by irreversibly inhibiting its protein degrading activity. TPT was shown to have greater potency in this regard than seven other organotins examined by the authors. The investigators deduced that the tin present in TPT interacts with the Nterminal threonine of the proteasomal β5 subunit, possibly provid ing a specific target for organotins. Organotins have long been known to induce necrosis; the authors propose this may occur through caspasedependent, DNA damage–independent cell death. In addi tion, the researchers assert that organotins most likely kill cells via a p53independent pathway. The new findings suggest that other previously identified poten tial targets of organotins, such as the transcription factor NFκB and the proapoptotic protein Bax, might be downstream of pro teasome inhibition. The investigators further posit that inhibition of aromatase activity observed in organotinexposed humans and animals—an effect linked to altered reproductive development— may be due to proteasome inhibition because such inhibition causes upregulation of factors that suppress transcription of the hCYP19/ aromatase gene. –M. Olfactory Transcriptional Analysis of Salmon Exposed to Mixtures of Chlorpyrifos and Malathion Reveal Novel Molecular Pathways of Neurobehavioral Injury.
Pacific salmon exposed to sublethal concentrations of organophosphate pesticides (OP) have impaired olfactory function that can lead to loss of behaviors that are essential for survival. These exposures often involve mixtures and can occur at levels below those which inhibit acetylcholinesterase (AChE). In this study, juvenile Coho salmon were exposed for 24 h to either 0.1, 0.5, or 2.5 ppb chl...
متن کاملThe Synergistic Toxicity of Pesticide Mixtures: Implications for Risk Assessment and the Conservation of Endangered Pacific Salmon
BACKGROUND Mixtures of organophosphate and carbamate pesticides are commonly detected in freshwater habitats that support threatened and endangered species of Pacific salmon (Oncorhynchus sp.). These pesticides inhibit the activity of acetylcholinesterase (AChE) and thus have potential to interfere with behaviors that may be essential for salmon survival. Although the effects of individual anti...

This review aims to provide an update on our current knowledge of the various effects of pesticide cocktails. We have collected data from studies conducted in mammalian models in vitro and in vivo that was published between 2000 and 2014. All ecotoxicological studies were voluntarily excluded. Cocktail effects were classified according to how they had been classified by each author. The frequen...

Two studies published recently looked into the health risks of organophospate and neonicotinoid plant protection products (PPP), upon the European honey bee (Apis mellifera L.) in Egypt. Using the concentrations reported in those studies a hazard assessment for consumption of Egyptian honey from the Nile delta was performed for both spring and summer pesticide exposure to humans. A total of 28 ...
متن کاملQuantifying Synergy: A Systematic Review of Mixture Toxicity Studies within Environmental Toxicology
Cocktail effects and synergistic interactions of chemicals in mixtures are an area of great concern to both the public and regulatory authorities. The main concern is whether some chemicals can enhance the effect of other chemicals, so that they jointly exert a larger effect than predicted. This phenomenon is called synergy. Here we present a review of the scientific literature on three main gr...
